What this test is for
Vitamin K1 (phylloquinone) is the main dietary form of vitamin K, found in green leafy vegetables like spinach, kale, and broccoli, and it works as a cofactor that activates proteins involved in blood clotting, bone strength, and inhibition of arterial calcification. This test measures the circulating level directly, which is most relevant when deficiency is suspected from poor intake or malabsorption (celiac disease, pancreatitis, chronic diarrhea, obstructive liver disease, or prolonged antibiotic use). Profound deficiency impairs the clotting factors made in the liver and causes excessive bleeding. Lower-grade insufficiency leaves bone and vascular proteins (osteocalcin, matrix Gla-protein) incompletely activated, which is why vitamin K status is studied in relation to bone density, fractures, and vascular calcification. Coumarin anticoagulants such as warfarin work by blocking vitamin K recycling, so dietary intake and these drugs interact directly.
