Prothrombin (Factor II) 20210G>A Genotype
An inherited mutation that raises the risk of abnormal blood clots in the veins.
Consider this test if:
- A personal history of unexplained deep vein thrombosis or pulmonary embolism, especially before age 50
- A first-degree relative with a known clotting disorder or recurrent blood clots
- Planning estrogen-containing contraception, hormone therapy, or pregnancy and weighing clotting risk
- A clot that occurred without an obvious trigger, or recurrent clots
- Already known to carry Factor V Leiden and clarifying combined inherited risk
- HSA/FSA eligible

Pre-test considerations
No fasting or special preparation required. This is a DNA test, so results are unaffected by recent meals, medications, or anticoagulant therapy, and timing does not matter. Anticoagulants do not change the genotype, though they do affect clotting tests like PT/INR.

What this test is for
The prothrombin G20210A mutation is one of the most common inherited causes of venous thrombosis, including deep vein thrombosis and pulmonary embolism. This test reads the F2 gene directly to determine whether you carry one copy (heterozygous) or two copies of the variant, which increases the production of clotting factor II. About 2.3% of the general population carries one copy, compared with roughly 6.2% of people with venous thrombosis and 18% of those with a strong family history of clots. The mutation rarely causes problems on its own; risk compounds with other factors such as Factor V Leiden, estrogen-containing contraceptives, pregnancy, surgery, immobility, and smoking.
This is a one-time genetic test, separate from prothrombin time (PT/INR), which measures how fast your blood is clotting right now, and from des-gamma-carboxy prothrombin, a liver and tumor marker. Because your genotype does not change, a single result settles it for life.

Frequently asked questions
View all FAQsHow does pricing work?
Every test shows the member price next to the standard non-member price, so you can see what membership saves you. The member price is our cost — covering the lab and what it takes to run the service — never a profit on the test itself; Mito makes its money on the $9 membership, not on marking up your tests. Membership is $9/mo, and you still pay the lab’s order fee. Prices are itemized before you pay, with no hidden fees.
Where do I get tested?
Choose a partner lab (Quest, Labcorp, or BioReference) at checkout. If your cart spans multiple tests, we consolidate the whole order onto a single lab so you only make one visit.
Is this eligible for HSA/FSA?
Yes. This test is HSA/FSA eligible, and you can pay with your HSA/FSA card at checkout.
When will I get my results?
Your results post to your dashboard once your lab completes them, then a clinician reviews them and your full analysis and personalized action plan (with clear next steps) follow. Turnaround varies by test: specialty assays and at-home kits take longer, and each test shows its expected turnaround before you buy.
Do I need a doctor’s order?
No. Mito provides the lab order for you, so you can book and get tested without a separate doctor visit.
