Pre-test considerations
Timing is the critical factor. For confirming ovulation, draw about 7 days after expected ovulation (around day 21 of a 28-day cycle, adjusted for your own cycle length). No fasting is required. Note any progesterone supplements or hormonal medication, since they affect the result.

What this test is for
Progesterone is the steroid hormone produced by the corpus luteum after ovulation, and by the placenta during pregnancy. It is barely detectable in the first half of the menstrual cycle, then rises sharply after ovulation to prepare the uterine lining for implantation, which makes a mid-luteal measurement the standard way to confirm that ovulation actually occurred. Low progesterone points to anovulation, a short luteal phase, or primary or secondary hypogonadism, while high levels appear in pregnancy and in certain forms of congenital adrenal hyperplasia. Because the level changes dramatically across the cycle, timing the draw correctly matters more than for almost any other hormone. In pregnancy it is also used to assess corpus luteum and placental function.

Where do I get tested?
